HGH Frag 176-191

HGH Frag 176-191 is the tail end of growth hormone — the last sixteen amino acids of a 191-residue hormone — sold on the idea that this stretch carries the fat-metabolising action and leaves the growth and blood-sugar effects behind. Two things complicate that. The published pharmacology was done on hGH 177-191, one residue shorter, and on AOD9604 and AOD9401, analogues from the same laboratory; AOD9604 swaps the leading phenylalanine for a tyrosine and is therefore a different molecule, which matters because AOD9604 is the one with a human trial history and a regulatory file. And every study of the unmodified fragment on record is a rodent study: obese mice and Zucker rats losing weight over two to four weeks, with lipogenesis suppressed in fat tissue. No human study of this peptide is on file, no half-life in any species is on file, and no dose in humans has ever been published.

Mechanism

The C-terminal domain of human growth hormone reduces lipogenesis and raises lipolytic activity in adipose tissue. In obese mice and Zucker rats the fragment inhibited acetyl-CoA carboxylase and stimulated hormone-sensitive lipase in the same direction as the intact hormone, and the analogue AOD9604 did so without binding the growth hormone receptor and without inducing cell proliferation, which is the basis for the claim that the lipid action separates from the growth and glucose actions. No receptor for the fragment has been identified.

Caveat on these identifiers

The FDA record on file concerns AOD-9604, a different molecule — hGH 177-191 with an extra N-terminal tyrosine — and not this fragment. In a December 2024 briefing document FDA concluded that a balancing of the criteria weighs against placing AOD-9604 free base or AOD-9604 acetate on the 503A bulks list, and found a lack of evidence for its effectiveness in obesity by any route. Nothing in that review evaluates hGH 176-191 itself.

Notes unverified prose

AOD9604 is a different molecule and its record should not be merged into this one. Drug Test Anal 2015 defines it as hGH 177-191 with an additional tyrosine at the N-terminus; that tyrosine replaces the phenylalanine at position 176, so the two peptides differ by a single hydroxyl (PubChem 71300630, C78H123N23O23S2, versus the free peptide of PubChem 172966176, C78H123N23O22S2). AOD9604 carries the human trial history and the FDA compounding review; hGH 176-191 carries neither.
